RESUMEN:  

Salamanders have the remarkable ability to regenerate their entire limbs
throughout life. Limb regeneration is an accurate process which gives
rise exclusively to the missing structures and in the correct
proportions, irrespective of the amputation level. This suggests that
cells in the stump have a memory of their position within the limb, a
property termed 'positional identity'. At present, very little is known
about how positional identity is encoded at the molecular level and how
it interplays with tissue growth. Through a single cell RNAseq approach
we recently identified the Tig1 tumour suppressor as a potential
determinant of proximal identity. Tig1 exhibits graded expression along
the proximodistal axis, is regulated by the proximalising factor
retinoic acid, and is able to change the fate of distal cells to more
proximal locations along the regenerating limb. In this talk, we will
discuss evidence supporting the role of Tig1 as a new determinant of
proximodistal identity and present molecular data that help to shed
light on its functions during limb regeneration.
